Which three-row Lexus is most efficient? The one that seats six.
The TX, the GX and the LX all offer seven seats. They are three very different vehicles, so you will be choosing on more than the seat count.
The thing to know first is that the TX 500h Performance Hybrid seats six, not seven, because it uses second-row captain's chairs. If you need seven, the TX 350 is your car.
And as with every three-row SUV, the cargo space to check is the one behind the third row, and you should measure it with your own bags.
The TX seats seven standard, and the GX and LX offer a third row on some grades.

The RX seats five as a two-row car. If a real third row is the goal, keep the TX, the GX and the LX on your list.
Will an adult sit in the back of it?
You are welcome to sit in the back yourself, with the middle row set where you normally keep it.

From there you can climb into the third row.
A few minutes seated back there answers most of the question.

Does the hybrid cost me a seat?
On the TX it can, and that is the first thing worth settling.

If you tow heavily or drive somewhere difficult, the GX is worth what it costs in fuel. If the daily school drive is the job, the TX does it on less fuel, and the hybrid on less again if six seats will do.

The difference is the second-row captain's chairs. They cost you the middle seat, so seven seats rules the hybrid out. On the TX you choose one or the other.
The fuel difference is four combined MPG, and you can have it only with six seats. If six is enough, take the hybrid: the captain's chairs also make the third row easier to reach.
If your seventh passenger is occasional, take the hybrid; the walkway to the third row matters more day to day than the fuel figure does. If seven is seven every day, the 350 is your car.
